Various components of natural allergens will stimulate the body to produce different specific IgE antibodies; such as house dust mites, more than 30 allergenic components have been identified and characterized, including tropomyosin of house dust mites, various Enzymes, regulatory proteins, structural proteins, etc.; such as milk, the main allergenic components are casein, α-lactalbumin, β-lactoglobulin, etc. These individual components are the allergen components.
Traditional diagnostic methods use crude allergen extracts, which measure the “sum” of IgE bound to all protein components in the entire allergen; component-analytical diagnostics use purified natural allergen or recombinant allergen components to identify allergenic Specific IgE, making the diagnosis of allergic diseases more precise.
Performance
Component allergens are purified by affinity chromatography and/or HPLC. Protein identity and amino acid composition are verified by mass spectrometry and by amino acid analysis. Immune reactivity is validated by monoclonal antibody binding. Purified allergens are also validated for IgE antibody binding.
